Distribution of estrogen receptors in subfractions of hen oviduct chromatin.

چکیده

Hen oviduct chromatin was digested with DNase II and separated into two fractions. The MgCl2 insoluble chromatin fraction (43% of the total DNA) was enriched in nucleosome-like particles, which sedimented at 11 S and contained 185 base pairs of DNA. The MgCl2 soluble chromatin fraction (5% of the total DNA) was characterized by 5 S and 14 S peaks in sucrose gradients; Estrogen receptors in the chromatin fractions were labelled with [3H] estradiol using the steroid exchange assay. The concentration of receptors in the MgCl2 soluble chromatin was 4;5 times higher than that in the MgCl2 insoluble chromatinmin sucrose gradient analysis the 11 S particles displayed a negligible specific radioactivity suggesting that estrogen receptors mainly bind to extranucleosomal chromatin.

Interaction of oestrogen and progesterone receptors with specific subfractions of laying-hen oviduct chromatin.

Salt (NaCl)-extracted nuclear oestrogen receptor from hen oviduct was incubated with salt-depleted oviduct chromatin and dialysed to low salt. The oestrogen receptor (re)associated with chromatin to form a 13-14S-sedimenting fraction, as found in 'native' chromatin, and saturation of this interaction was obtained for very low receptor concentrations (approx. 0.04 nM). Similarly, purified proges...

Estrogen receptor in hen oviduct chromatin, digested by micrococcal nuclease.

Nuclei from laying hen oviduct were prepared according to Hewish and Burgoyne i.e. in the presence of spermine and spermidine and in the absence of divalent cations and were then moderately digested by micrococcal nuclease. When the resulting chromatin was analysed by ultracentrifugation on a sucrose gradient, a peak of specific estradiol-binding sites was observed, sedimenting slightly faster ...

Isolation of estrogen receptor in complex with a discrete nuclear subfraction from hen oviduct.

A nuclear subfraction containing bound estrogen receptor in presumed complex with its nuclear acceptor site has been partially purified from hen oviduct. Sucrose density gradient ultracentrifugation was used to separate mechanically sheared chromatin (i.e. lysed nuclei) into several fractions which differed in protein to DNA ratio as well as in vitro template activity. Gradient fractions were t...

Evidence for specific DNA sequences in the nuclear acceptor sites of the avian oviduct progesterone receptor.

Recent studies have shown that saturable high-capacity nuclear binding sites (termed acceptor sites) for the avian oviduct progesterone receptor can be reconstituted by rehybridizing a specific oviduct chromatin protein fraction (CP-3) to pure hen DNA to generate a reconstituted nucleoacidic protein (NAP). Only a limited number of acceptor sites can be generated on hen DNA even at high protein/...

Effect of Estrogen on Gene Expression in the Chick Oviduct COMPARATIVE ASPECTS OF RNA CHAIN INITIATION IN CHROMATIN USING HOMOLOGOUS VERSUS ESCHERICHIA COLZ RNA POLYMERASE*

Hen oviduct RNA polymerase II and Escherichia coli RNA polymerase holoenzyme and core enzyme were used to study the initiation of RNA synthesis on chromatin. In either the presence or absence of estrogenic stimulation, changes in the level of oviduct chromatin initiation sites as measured in the presence of either homologous or heterologous polymerases followed a similar pattern. Comparison of ...
